Abstract
Structural analogues of chloramphenicol (CAP) cause mechanism-based inactivation of rat liver cytochrome P-450 (P450) either via protein acylation or destruction of the heme prosthetic group. The first objective of this thesis was to determine whether CAP analogues that cause loss of the P450 heme moiety also cause porphyrin accumulation in chick embryo liver cell culture. The second objective was to determine uroporphyrinogen decarboxylase (UROG-D) activity in 17-day old chick embryo liver homogenates by measuring the conversion of pentacarboxyl-porphyrinogen I to coproporphyrinogen I (pH 6.0). The third objective was to estimate the time of UROG-D inhibitor formation by concurrently adding ALA (15$\mu$g/mL) and nifedipine (25$\mu$g/mL) or TCBP (2$\mu$g/mL) to chick hepatocyte culture.
